چکیده
Abstract Background ADP/ATP translocase 1 (ANT1) is involved in the exchange of cytosolic ADP and mitochondrial ATP, its defection plays an important role pathogenesis. To reveal etiological implication ANT1 for Parkinson’s disease (PD), a neurodegenerative disorder, mouse model treated with 1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine neuroblastoma cell induced by 1-methyl-4-pehny1-pyridine were utilized this study. Results The tissue-specific abundance brains was accessed using analysis Western blot immunohistochemistry. Down-regulated soluble found to be correlated PD, associated PD pathogenesis via forming protein aggregates α-synuclein. This finding confirmed at cellular level models. supplement neuronal cells revealed protective roles against cytotoxicity caused MPP + . Protein interaction assay, coupled LC-MS/MS, silver-stained SDS-PAGE anti-ANT1 antibody respectively, illustrated α-synuclein expressed as bite. Additionally, significant increasing ROSs detected -treated cells. Conclusions study indicated that potentially causative factor led neuropathogenic injury promoting formation investigation promotes innovative understanding on etiology provides valuable information developing potential drug targets treatment or reliable biomarkers prognostication.

Anti-Human α-Synuclein N-Terminal Peptide Antibody Protects against Dopaminergic Cell Death and Ameliorates Behavioral Deficits in an AAV-α-Synuclein Rat Model of Parkinson’s Disease
The protein α-synuclein (α-Syn) has a central role in the pathogenesis of Parkinson's disease (PD) and immunotherapeutic approaches targeting this molecule have shown promising results. In this study, novel antibodies were generated against specific peptides from full length human α-Syn and evaluated for effectiveness in ameliorating α-Syn-induced cell death and behavioral deficits in an AAV-α-...

As our society ages, neurodegenerative disorders like Parkinson`s disease (PD) are increasing in pandemic proportions. While mechanistic understanding of PD is advancing, a treatment with well tolerable drugs is still elusive. Here, we show that administration of the naturally occurring polyamine spermidine, which declines continuously during aging in various species, alleviates a series of PD-...

MicroRNAs (miRNAs) are short, 22–25 nucleotide long transcripts that may suppress entire signaling pathways by interacting with the 3’-untranslated region (3’-UTR) of coding mRNA targets, interrupting translation and inducing degradation of these targets. The long 3’-UTRs of brain transcripts compared to other tissues predict important roles for brain miRNAs. Supporting this notion, we found th...
متن کاملEndosulfine-alpha inhibits membrane-induced α-synuclein aggregation and protects against α-synuclein neurotoxicity
Neuropathological and genetic findings suggest that the presynaptic protein α-synuclein (aSyn) is involved in the pathogenesis of synucleinopathy disorders, including Parkinson's disease (PD), dementia with Lewy bodies (DLB) and multiple system atrophy. Evidence suggests that the self-assembly of aSyn conformers bound to phospholipid membranes in an aggregation-prone state plays a key role in a...
متن کاملSIRT1 protects against α-synuclein aggregation by activating molecular chaperones.
α-Synuclein is a key molecule in the pathogenesis of synucleinopathy including dementia with Lewy bodies, Parkinson's disease, and multiple system atrophy. Sirtuins are NAD(+)-dependent protein deacetylases that are highly conserved and counter aging in lower organisms. We show that the life span of a mouse model with A53T α-synuclein mutation is increased by overexpressing SIRT1 and decreased ...
